Microsatellite assay for dog parentage testing and identification
Canine Genotypes Panel 1.1 is a reagent kit for dog parentage testing and identification using microsatellite (STR) DNA loci. The kit allows co-amplification of the microsatellites in a single multiplex PCR reaction.
The Canine Genotypes™ Panel 1.1 kit encompasses the following 19 loci: AHTk211, CXX279, REN169O18, INU055, REN54P11, INRA21, AHT137, REN169D01, AHTh260, AHTk253, INU005, INU030, Amelogenin, FH2848, AHT121, FH2054, REN162C04, AHTh171 and REN247M23. These markers are included in the 'core panel' of loci recommended by the Applied Genetics Committee of Companying Animals of the International Society for Animal Genetics (ISAG).
Reagents and protocols of the Canine Genotypes™ Panel 1.1. kit have been optimized to deliver similar peak sizes (amplification yields) for each loci. The kit employs Phusion Hot Start DNA Polymerase. Allele callings obtained with this kit represent the true alleles of an individual, instead of 'plus-A peaks' or 'split peaks' typically interpreted when using e.g. a conventional Taq DNA polymerase. This is due to the proofreading activity of the Phusion Hot Start Polymerase.
The Canine Genotypes Panel 1.1 kit allows co-amplification of the 19 markers
in a single multiplex PCR reaction. One primer from each primer pair is end-labeled
with a fluorescent dye. After PCR, the fragments are separated and detected
in a single electrophoresis injection, using an automated electrophoresis instrument (Figure 1).
The Canine Genotypes Panel 1.1. kit provides all the necessary reagents
for amplification of the 19 microsatellite loci. In addition, the kit includes
canine control DNA originating from an ATCC cell line for verifying acceptable PCR and electrophoresis conditions.
