DNA amplification with real-time PCR technology

In the PathoProof Mastitis PCR Assay, the mastitis causing bacteria are identified by amplifying the bacterial DNA, not by culturing bacteria on plates. Whereas culturing methods can reveal the presence of viable bacteria only, DNA amplification technology allows identification of viable, dead, and also growth-inhibited bacteria. This decreases the number of “no growth” results, a problem occurring with the conventional bacterial culture method.

The DNA amplification technology used with the PathoProof Mastitis PCR Assay is real-time PCR. This technology amplifies DNA by PCR (polymerase chain reaction) simultaneously identifying the amplified DNA fragments in “real time”. In contrast to all other PCR technologies, real-time PCR is a fully automated, closed-tube system having important benefits for the laboratory, such as:

  1. Elimination of laboratory contamination risks. Amplified PCR products are the primary source for cross-contamination in a laboratory running PCR assays. In real-time PCR, the reaction vessels are not opened, which eliminates the risk of contamination.
  2. Automation and throughput. All other DNA technologies require complex processes after the PCR reactions have been done. The real-time PCR process is fully automated, decreasing the possibility of human errors and increasing laboratory throughput. 
  3. Quantification of DNA. Real-time PCR is the most accurate quantitative DNA method available.

Use of the PathoProof Mastitis PCR Assay requires a real-time PCR instrument. Finnzymes collaborates with Agilent Technologies as a global distributor of the Stratagene Mx3005P® QPCR System to be supplied with the kit.
